High-performance Frontal Analysis, HPFA)法は HPLCシステムを利用する薬物 血漿タンパク結合分析法であり、オンラインで簡便、高感度な結合分析が可能となる。これまでの検討では、血中モデルタンパク質としてヒト血清アルブミン( HSA)を採用し、本法にて様々な薬物との解析を試みてきた。HSAは 分子量約66500の主要な血漿タンパク質であり、通常35~50g/L≒ 500~750μMの濃度で血漿中に存在しているが、モデル試料においてはHPFA解析モデルの立証と再現性のみに注視し、100μMの濃度で固定して測定を行ってきた。そこで本研究では、試料中HSA総濃度を 変更したときに、薬物-血漿タンパク結合解析から得られる結合定数に変化が見られるかを検討した。オンラインHPFAシステムを構築し、HPFA分離カラムにDevelosil 100Diol-5(6.0 mmI.D.×150mm, Nomura Chemical Co.,Ltd)、薬物定量カラムに5C18-MS-II(2.0mmI.D.×150mm,nacalai tesque,Inc.)を使用、モデル薬物にはphenylbutazone(PB)を 採用し、100、50、10μMのHSA総濃度に対し複数のPB濃度で混合した試料を作成して、カラム温度37℃の条件で測定した。得られた非結合型薬物濃度からLangmuir型のプロットより回帰計算(Kaleida Graph™ ver.4.5, Synergy Software)を行い、結合定数を算出した。HSA総濃度を変化させてもLangmuir型プロットは同一のものとなり、算出した結合定数、及び結合サイト数も同一の値となった。本検討結果より、HPFA解析においてHSA濃度を変化させても影響はなく、薬物
