| 内容記述 | The present study aimed at examining the embryolethality and teratogenic potential by 6-aminonicotinamide (6-AN), and determining the critical period following a single administration during the organogenesis in mice. 6-AN at 15 mg/kg was administered orally to females either on gestation day (GD) 8, 9, 10, 11, 12, 13 or 14, and cesarean section was conducted on GD 18. The uterine implants were classified as live fetus, or resorbed or dead fetus. The live fetuses were weighed and examined for external or skeletal anomalies. There were significant or slight increases in the number of resorptions, dead fetuses or % of postimplantation losses and the corresponding decrease in the number of live fetuses, and significant decreases in live fetal weights in any treatment groups. In the external examination, cleft palate was mainly observed in the GD 9, 10, 11, 12, 13 or 14 treatment groups, and club foot or hindlimb digital anomalies were observed in the GD 9 or 10 treatment groups. In the skeletal examination, the vertebral or hindlimb anomalies were observed in the GD 8, 9 or 10 treatment groups. In conclusion, a single oral administration of 6-AN at 15 mg/kg during the organogenesis clearly revealed that embryolethality and teratogenic potentials of cleft palate, vertebral or hindlimb anomalies, and the critical period of these anomalies were determined GD 9 to 14, GD 8 to 10, or GD 9 to 10, respectively, in this study condition. | |||||
